CYP154C5 from Nocardia farcinica is a P450 monooxygenase able to hydroxylate a range of steroids with high regio- and stereoselectivity at the 16α-position. Using protein engineering and substrate modifications based on the crystal structure of CYP154C5, an altered regioselectivity of the enzyme in steroid hydroxylation had been achieved. Thus, conversion of progesterone by mutant CYP154C5 F92A resulted in formation of the corresponding 21-hydroxylated product 11-deoxycorticosterone in addition to 16α-hydroxylation. Using MD simulation, this altered regioselectivity appeared to result from an alternative binding mode of the steroid in the active site of mutant F92A. MD simulation further suggested that the entrance of water to the active site caused higher uncoupling in this mutant. Moreover, exclusive 15α-hydroxylation was observed for wild-type CYP154C5 in the conversion of 5α-androstan-3-one, lacking an oxy-functional group at C17. Overall, our data give valuable insight into the structure–function relationship of this cytochrome P450 monooxygenase for steroid hydroxylation.  相似文献

In recent years, the finite element method (FEM) has become the main tool for simulating the metal cutting process because research based on trial and error is time consuming and requires high investment. Early studies were done by different investigators. In this research AISI 52100, hardened steel (62 HRC) was selected for an orthogonal machining process as well as metal cutting simulation using the software DEFORM-2D. This software is based on a forging process and has been adapted to an orthogonal machining process. The results of simulated cutting forces were compared with experimental cutting force data to validate the orthogonal cut simulation. Also, the surface roughness was measured, and the influence of the stress, strain, and temperature on the surface roughness was studied.  相似文献

Samples of mesoporous silica SBA-15 were prepared under hydrothermal conditions where Cu cations were incorporated to the structure by impregnation in order to compare the adsorption behavior in the presence and absence of this element. The adsorption/desorption equilibrium isotherms of propylene, propane, and N2 were measured to evaluate their usefulness in the propane/propylene separation. All the adsorption isotherms of SBA-15 measured in the absence of Cu cations were described by the Freundlich equation, while the adsorption isotherms of propane on Cu/SBA-15 were better represented by the Henry equation and those of propylene were satisfactorily described by the Langmuir model in the range P < 100 Torr. The adsorption uptake of propylene increased and that of propane decreased in Cu/SBA-15 as compared to the amounts observed in the SBA-15 sample. The presence of Cu atoms in the adsorbent lattice increased the selectivity towards propylene. Under some working conditions the adsorbed amount of propylene in Cu/SBA-15 sample was totally reversible and the propane uptake, negligible.  相似文献

Gutta‐percha cone is the most widely used material for root canal filling. The in vivo aging of this cone focus on the degradation of its main organic component, trans‐1,4‐polyisoprene, was studied. Aged cones (25 samples) from 2 to 30 years of root canal filling were extracted from different patients in the occasion of retreatment by mechanical way. The information about the aging time was given by the patients. Gel‐permeation chromatography (GPC) and infrared spectroscopy (FTIR) were the analytical techniques used. Polyisoprene degrades with time of aging, but in a slow process. Decrease in polymer molar mass from 5.7 × 105 to 1.7 × 105 g/mol was observed in polyisoprene from cone after 30 years of root canal filling and inside a noninfected tooth. In tooth with caries and periodontal infection, the decrease in molar mass is higher (4.6 × 104 g/mol in cone with 10 years of aging). The production of carbonyl and hydroxyl groups in the aged material indicates that the process is oxidative, even in closed teeth. In these cases, the oxygen could be provided from tissue fluid. The degradation mechanism is complex and depends on many factors, besides time of root canal filling. The dental problem caused by the aging could be the production and migration of cytotoxic substances to periodontal ligament and the reduction on the canal sealing property due to the polymer weight loss. Summary Copolymeric poly(acrylic acid-co-methyl methacrylate) hydrogels for three different compositions: (90/10), (80/20) and (60/40), have been studied. Drug release has been examined as a function of the hydrogel composition by HPLC (High Pressure Liquid Cromatography). The release experiments were carried out at 37 °C. The fraction of available drug release was linear in t1/2. The values of the diffusional coefficient (0.50<n<1.0) indicate that the nafcillin release mechanism from the hydrogels in study is non-Fickian. The diffusion coefficients for this drug release have been calculated. The molecular diffusion of nafcillin through hydrogels is controlled by the swelling.  相似文献

A series of semi-interpenetrating polymer networks (semi-IPN's) based on different compositions of an acyclic polyethylene terephthalate oligomer and unsaturated polyester resin (UP) were prepared. The oligomer was dissolved in the resin containing styrene crosslinker. Later this mixture was crosslinked at room temperature using methyl ethyl ketone peroxide catalyst and cobalt naphtanate as promoter. The tensile strength of the IPN's decreases as the concentration of oligomer increases, whereas, elongation to break increases. A characterization of the oligomer used is also presented.  相似文献

Chemo-enzymatic synthesis of amino acid-based surfactants   总被引:5,自引:0,他引:5  
The application of lipases to the synthesis of amino acid-based surfactants was investigated. Low yields (2–9%) were obtained in the acylation of free amino acids, such as l-serine and l-lysine, as well as their ethyl esters and amides with fatty acids, owing in part to low miscibility of the reactants. When the N-carbobenzyloxy (Cbz)-l-amino acids were used in an effort to improve miscibility of the amino acid derivatives with the acyl donor, a dramatic improvement was observed for N-Cbz-l-serine (92% yield) but not for N α-Cbz- or N ζ-Cbz-l-lysine (7 and 2% yield, respectively). As an alternative, and efficient synthesis of N ζ-acyl-l-lysines was developed, based on the regiospecific chemical acylation of copper(II) lysinate. In pursuit of a general route to amino acid-fatty acid surfactants, the utility of a polyol linker was investigated. Thus, the glycerol ester of N α′ N ζ-di-Cbz-l-lysine was prepared and evaluated as a substrate for acylation. As expected, this and other glycer-1-yl esters of N-protected amino acids were excellent substrates for lipase-catalyzed acylation. Their reaction with myristic acid in the presence of Novozyme resulted in the regioselective acylation of the primary hydroxyl group of the glycerol moiety to afford the corresponding 1-O-(N-Cbz-l-aminoacyl)-3-O-myris-toylglycerols with conversions of 50–90%. These were readily deprotected to give a range of 1-O-(aminoacyl)-3-O-myristoyl-glycerols with overall yields of 27–71%.  相似文献

The series of nylon 612 copolymers was synthesized from caprolactam (C) and laurolactam (L) at 145°C. The 50/50 C/L molar ratio copolymer was found to have the minimum melting temperature (Tm ) for the series. The glass transition temperatures (Tg 's) of the copolymers were affected by the crystallinity of the copolymers. The Tg was at a minimum for the 50/50 copolymer for crystalline samples. However, for amorphous samples there was a decrease in Tg with increasing L content. Percent crystallinity was determined by differential scanning calorimetry and X-ray techniques. It was found that the degree of crystallinity was at a minimum for copolymers of 70/30 to 40/60 C/L ratios. Coefficients of linear thermal expansion (CLTE) were obtained for the copolymers at 10°C intervals from 20 to 70°C for dry and from 20 to 50°C for samples conditioned at 50% relative humidity and 50°C. The dry samples gave lower initial values, but had a greater temperature dependence than the conditioned samples. As expected, the CLTE was found to be lowest for samples exhibiting the highest crystallinity. The tensile strengths and moduli decreased rapidly with increasing L up to the 70/30 C/L ratio after which they remained relatively constant. Elongations reached maximums between 70/30 and 40/60 C/L ratios. An inverse relationship was found between crystallinity and impact strength.  相似文献

The results of the chemical composition and biological quality of deffated hazel nut flour are shown. The samples analyzed contained significant amounts of proteins (19%) comparable to legume flour, higher than cereals and lower than deffated oleaginous flours. The oil extracted from the seed was analyzed and the average results obtained were the following: Refraction index, 1.47; saponification No. 184.8; iodine No. 85.0. The average composition of the fatty acids obtained by gas liquid chromatography was: Palmitic acid 2.3% Palmitoleic acid 37.0% Stearic acid 0.5% Oleic acid 39.5% Linoleic acid 6.9% Linolenic acid 1.1% Eicosanoic acid 2.3% Eicosaenoic acid 4.6% Docosenoic acid 3.4% Tetraeicosanoic acid 0.3% These results indicate a good-quality oil due to the low content of linolenic acid. The nutritive value of the deffated meal measured in the rats gave a net protein ratio (NPR) of 3.58, lower than the corresponding casein value (4.10). The true protein digestibility measured in the rat gave a value of 7.3%, compared to 95% for casein. The amounts of iron and phosphorous are comparatively lower than those reported for rape-seed meal and sunflower meal.  相似文献

In this paper, an asymptotic expansion is constructed to solve second-order differential equation systems with highly oscillatory forcing terms involving multiple frequencies. An asymptotic expansion is derived in inverse of powers of the oscillatory parameter and its truncation results in a very effective method of dicretizing the differential equation system in question. Numerical experiments illustrate the effectiveness of the asymptotic method in contrast to the standard Runge–Kutta method.  相似文献
